Are activated proto-onc genes cancer genes?

چکیده
The main objective of cancer molecu~ar biology is to identity cancer genes. ~espite fierce efforts, this objective has still not been met [1-3]. As yet the only known cancer genes are the transformin~ one g~n.e~ of retroviruses. Typically these VIruses InItiate and maintain cancers with autonomous transforming genes that ~re domina~t in susceptible cells [5]. The dIsc~very of ~Ingle gene determinants of cancer In retroVIruses has become a precedent that has infected cancer gene research. It has made retroviral one genes the favorite models of cellular oncogenes, although th~ releva~ce of single-gene models to VIrus-negative tumors is as yet unknown. Fortunately, one genes are either detrimental or at least useless to the viability of the virus and thus are not maintained by retroviruses. They are the products of rare, genetic ac.cid~nts, generated by illegitimate recombmatIOns between retroviruses and cellular genes,

Cancer genes: rare recombinants instead of activated oncogenes (a review).
The 20 known transforming (onc) genes of retroviruses are defined by sequences that are transduced from cellular genes termed protooncogenes or cellular oncogenes. Based on these sequences, viral onc genes have been postulated to be transduced cellular cancer genes, and proto-onc genes have been postulated to be latent cancer genes that can be activated from within the cell to cause virus-negat...

The transforming (onc) genes of oncogenic retroviruses share most or all of their coding sequences with normal cellular genes termed proto-onc genes. The viral genes differ from proto-onc genes in virus-specific promoters and in various point mutations and substitutions of cell-derived coding regions. In view of the structural similarities between viral oncogenes and cellular proto-onc genes, t...

The oncogene hypothesis postulates that mutated cellular genes, termed proto-onc genes, function as cancer genes because they are related to retroviral onc genes. However, in contrast to retroviral onc genes, mutated proto-onc genes from cancers are not sufficient for carcinogenesis. Therefore, it has been proposed that mutated proto-onc genes depend on other proto-onc genes for carcinogenesis....
متن کاملViral oncogenes and cellular prototypes.
The structural hallmark of retroviral transforming onc genes is a specific RNA sequence that is unrelated to the essential retroviral genes but closely related to certain cellular prototypes termed proto-onc genes. Two types of onc genes have been distinguished. Type I are onc genes which utilize elements of specific sequences only to encode a transforming protein. Type I1 onc genes are hybrids...
